What is tree root DNA testing?

Molecular analysis of tree roots has the potential to identify individual trees, reducing costs and the risk of insurers. The method of DNA testing is based on the use of specific sequence repeat markers and enables the identification of the root material at species level.

Traditionally, the methods used for root study were limited by low precision and low accuracy. The methods used for identifying tree roots include microscopy and comparison with root anatomy manuals.

In the past decade, DNA-based techniques have been implemented to identify root species. However, there is no consensus on the best method of root study. This is due to the lack of reference samples.

ISSR primers can distinguish different individuals of a species, which makes them an effective tool for invasive tree root identification. In addition, the technology has high repeatability and offers sufficient variability to detect genetic variation among individuals closely related to each other.

Tree root ID may be possible through improved technologies based on wood anatomy and tree species’ DNA. In addition, tree root identification may be useful in determining whether the tree roots found in trial pits are the same species.
